Is the NCAA going to have 32 sites for first round games, and if they are, how will that impact potential brackets especially regarding first round match-ups?

Yes, thirty-two teams will be hosting first round games. To answer your other question, nobody really knows, because the powers that be have been hilariously vague about how they’ll be setting up the bracket with the new format.

Will this mean that the higher-seeded team will be hosting each match? In the past, with 16 sites, they could screw over the highest-seeded team, citing the combined travel burden on the other three teams as a rationalization. With only two teams involved, it is a zero-sum game whether A travels to B, or vice versa. (There could still be exceptions made for cases in which, say, the higher-seeded team is unable to provide a suitable venue.)
